Promega has developed a new fast immunoassay platform (Lumit™ immunoassays) for quantifying targets, such as cytokines (Lumit™ cytokine immunoassays), without the need for any wash steps.
The technology allows rapid and sensitive determination of cytokine release from cells without sample transfer. All you need to do is add labeled antibodies, wait one hour, then add the detection substrate and read the assay plate on a standard luminometer.
In Lumit™ cytokine Immunoassays, antibodies have been chemically labeled with the small and large subunits of NanoLuc® Luciferase, known as small bit (SmBiT) and large bit (LgBiT), respectively. In the presence of an analyte, the two antibodies come into close proximity, allowing SmBiT and LgBiT to form an active enzyme and produce luminescent signal proportional to the amount of cytokine present.
The Lumit cytokine platform allows for faster and easier detection with different ready-to-use detection kits (IL-1B, IL-2, IL-4, IL-6, IL-10, TNFα, IFNγ,…), supporting researchers interested in understanding the cytokine storm, looking for modulators of the inflammasome, and other inflammation and immunity research. Next to the cytokine assays, we are also able to provide several types of Lumit™ Immunoassays, including specific kits to detect protein phosphorylation, metabolites or neonatal Fc receptor binding.

Applied innovations:
Traditionally, ELISAs have been used for cytokine detection, but the process is often tedious and they can take up to 24 hours to generate results. With the new Lumit™ cytokine immunoassays, users can detect released cytokines directly in-well with cultured cells. With zero wash steps, you can get results in 70 minutes or less.
These assays share excellent sensitivities (LLODs <10pg/ml) and broad linear ranges (>3 logs of cytokine concentration), significantly mitigating the need for sample dilution.
Because of these advantages, LUMITTM immunoassays are easily adaptable to automation for labs processing large numbers of samples.
